our first looking is simply the interaction between secretary i ga this is

uh looking a milk circuitry i. j. so a very heterogeneous group of this

like a protein interaction with select potential uh select
00:05:51
commands alls and looking and it's increase in conversation potential

so i've investigated the digestion of these mode of these onion complexes secretary

and you come and so complexes adult in beach oh and in the val

and the ability for this to improve the attachments both again in in future systems and also in a mouse model

then investigating intolerance in the host and second looking at the complex itself
00:06:16
so are there any specific functions of this commands all secretary i ga complex

and the some looking at compensation resistance so potentially the

ability for this um this complex to help reduce pathogen binding

um perhaps in improved pathogen clearance and of course have improved barrier function on the host

and this knowledge uh that we are gaining with the study is not only

will help us investigate mechanisms as to what's happening in natural breast milk system

but it surely in the development of a product that may improve

the ability for probing attics um well it may improve administration probably addict

this is looking at a possibility of creating some complex perhaps with remote carcass

like the solace differed a bacterium some very common commands of bacteria of the infant

that when administered to the infant may help protect the probing

so i was looking at these as a currently marketed um probably attics

some of these questions initially looking at the interaction between secretary actually increments alls

first is their concentration dependent binding of secretary i do to commence of bacteria

and second how important are these and went quite cans so does the removal

of these like ants lead to a reduction in the association of secretary i ga

question uh to an extent and so part of this was we could be

cheerleading his studies and the other part of this was really as a novel investigation

and so what i found uh and i use flow sitcom entry to analysts much of my secretary

and uh i used a um a anybody actually that jeff gordon used also in study this is

a lexus six forty five detection antibody for secretary i ga this is an example of a flow plot

and uh hopefully you can see a very clear demonstration haven't increased association of my um bacteria

certainly evidence that this uh interaction is concentration dependent

and i wanted to look at this a little bit more detail with some of the top binders

so like to solace from this is that o. g. g. guy um beyond the mongol maybe longman finances all three had

very robust binding under the highest physiological concentrations of secretary i ga
00:11:07
and when i used a variations of this concentration you can see that
00:11:12
uh these organisms performance a little bit differently well with their
00:11:15
interactions with secretary i ga but interesting link all of them
00:11:18
seem to reaches saturation and then type so interesting to me and perhaps those are you in the breast milk world because
00:11:25
again the physiologically relevant concentration a secretary i ga in breast milk
00:11:30
it's about one meg per mail for this uh perhaps demonstrates that in

that press they'll knock environments we actually have low word saturation of our commence

a bacteria that are found in a press not community and then then

that they could be reaching 'em but more investigation is a record for this

to answer the second part of this question how important are these and link quite cans at this interaction

and uh oh let's get these in one has really study showed that the secretary component alone

was protective with the digestion of the idea molecule and so with this i hypothesise that perhaps

commence a bacteria coded with the secretary coded i. g.

and so i developed a model and kind of take a lot of trial and error to um

to evaluate this question surpass bacteria two times a second time with the on the sugar substrate of interest

i because for all bacteria lactose and then that preferred each most substrates for some buffer bacteria

uh i checked the associations right i a call
00:13:40
to these bacteria bounded with secretary i ga not right
00:13:44
check the association with that fax came and then with this complex here
00:13:49
i then subjected and this complex to i gastric digestion
00:13:53
conditions using p. h. of four with percent perhaps and
00:13:56
and then uh um intestinal indigestion with pink retain at each of seven
00:14:01
with this complex i checked balls uh live counting with see if you um yes you're dilution and see if
00:14:06
you counting and then i also used to uh flows for said how much she with a set of toxicity essay
00:14:12
and i evaluate this with with all organisms have interests in found variations in in
00:14:18
uh the protective capacity of secretary i j. but under all circumstances with every bacteria lines
00:14:24
there did seem to be protective impact uh using secretary i ga so this is
00:14:28
lawful or fold change over and these organisms digested but out that protected in a body
00:14:35
in some cases every statistically significant uh and other cases very close to but um will be
00:14:41
repeating somebody's ass is to truly determine this that in all cases of i did a regression fine
00:14:45
uh using all different um concentrations the secretary a
00:14:48
g. a. i certainly some uh i demonstrated correlation behind
00:14:52
and the protective ability for secretary i ga to increase viability of bacteria postage action

so this was an in beecher model and uh i wanted to see if this holds true in a mouse model

and so on this for study this is a a um an investigation using biopsy mice

this trial we were added a very short trial to investigate the survival uh and i choose to use the long gone

a long concerts pieces in finances and again this is one of these champion connoisseurs of the infant got so

surely trying to um tease out what's happening uh in as best as i could with these in think human souls
00:15:30
for this trial by a collective baseline samples here to determine uh into that
00:15:34
there were no will be in fantasy since really no better bacterium in these mice
00:15:38
i'll buy three days i've devices with the intent is either

so she did or not associated with not secretary i ga

i use that concentration i had found that was
00:15:48
the um highest saturation rate for this particular bacteria

and then collected people samples one they pose to garage to conceive on the second day

and to determine the amount of bacteria that essentially were able to survive

that passage and and persist for at least a short period of time

and what i found was indeed that when the intent is was pretty associated with secretary i

ga and this was knock i ga and that i had almost a complete lock fold increase
00:16:17
of bacteria that we're able to passage through that and uh it has no content and um be sure to the feces
00:16:23
now i will make one caviar that was that one of my colleagues brought to
00:16:26
my attention this is fickle bacteria so i i cannot say with this plot alone that
00:16:32
this demonstrates persistence in the intestine we do have buyer geography studies that certainly do show
00:16:38
there is a still also same not quite about ten to that but see if you
00:16:43
in almost every part of the intestine um post um post ingestion so at that day evan across the
00:16:50
uh so i can i completely say this is due to just shedding of the bacteria

uh or if it's evidence of survival of the bacteria but um but nonetheless again with by geography studies i can say that

these little still did seem to be much higher in those um mice

that were provided that the influences that was pretty associated with a. g. m.
00:17:12
so certainly at least in the study here we do do see some evidence that secretary

i. g. a. can improve that survival of commands a bacteria postage russian induced obscene mice

and the last part of this initial investigation we just commences and secretary i ga

was to look at this ability for um these commences to bind to build an inbuilt show 'em

so for the beecher trials i uh against humans trial and error i came

yeah for these ones i can use the in fantasy and i wanted to just do an evaluation of

persisted be in fantasy after a short period of time sitting on

these coincides and look at any expression changes in the clan sites themselves

and again what i found was as i increase the association in this is

pretty association before i put the bacteria on to the surface these cullen inside

so as i increase that percentage of population associated with the
00:18:24
infinite jest i also so i've very clear um increase in
00:18:28
the amount of bacteria there remain down to the surface of
00:18:31
these concise even after watching uh many times with p. b. s.

and just a plot out to to that sloping give you clear number so for

every one percent increase association for the spectre population you see about ten to the fourth

see if you increase binding capacity so seem to be a very robust um binding

capacity with secretary i. g. m. and then again gene expression studies on these co cultures

when the intent as was provided in this is like fold or full changeover baseline with p. b. s. alone

there's a significant alteration a number of different genes involved

when the mouse was provided be in fantasy with that too because uh lactose allow

we now we're able to detect levels of persistence or all all these this trial

so in this case it looks like the uh that too few cars selectors alone will increase persistence of this the influences

now this is also true if you've been following any of the most work uh

we've also looked at this but you want them longer messy five nine six and found

the act essentially the functional cross reactivity of a monoclonal recombinant

and this is uh looking at those that were able to invade cullen slides or

to migrate to the priors patches in a mouse model and the idea here are

competitive index of one essentially means an equal number of all the while type and onions

um and a combat index anything less than one indicate self

or specificity where we had a fewer number of the wild type

and then no change in the mutant so we're looking at this as

our um as a baseline markers to evaluate effectiveness of self or specificity

and also to get away from the inherent variability in mouse models and to look at seminal an invasion

parent teacher models we use again we actually for both we use the in fantasy

and after treatment groups we have four groups here first

is the same analysis or positive control just seminal alone
